1) Injectable L-Carnitine
You’ve probably heard of L-Carnitine. It’s just a boring amino acid right?
Wrong.
It’s quickly becoming one of my favorite nutrients of all-time the more I use it. This became even more apparent when I switched over from L-Carnitine L-Tartrate & ALCAR to Injectable L-Carnitine. More on why I did that later.
The #1 reason why I use it?
It turns fat into fuel.
More specifically, it works by transporting long-chain fatty acids into the mitochondria to be burned for energy by a process called β-oxidation. R
And because of this, it’s been proven in human studies to enhance weight loss, even when taken orally. R R
L-Carnitine’s fat-burning effects are even more prominent when used in conjunction with medium to low exertion cardio through its lipid oxidisation and glycogen sparing abilities R
Additional Relevant L-Carnitine Benefits
Androgenic Activity: L-Carnitine intake has also been shown to upregulate androgen receptors, which basically makes your testosterone work harder for you. All of the androgenic benefits you normally get from testosterone will be enhanced while using taking L-carnitine, including a massive benefit sexual performance. It’s so powerful that it has been called a natural alternative to TRT. And in a human trial, L-Carnitine actually outperformed TRT in some markers of sexual function without significantly altering any sex hormone levels. When I started taking higher doses of L-Carnitine, I definitely noticed improved sexual health as well (stronger erections & sex drive mainly). Improved androgen signaling will also favorably benefit body composition. R R
Increases IGF-1 levels: IGF-1 is the anabolic hormone responsible for many of the hypertrophic benefits of growth hormone. R
Increases ATP production: By turning fat into fuel, L-Carnitine also increases mitochondrial ATP energy output, resulting in a noticeable energy boost which is especially useful for workouts. R
Enhances Muscle Recovery: L-Carnitine has been shown to improve blood flow, oxygen, & glycogen levels in muscles. This is in part why it expedites muscle recovery and reducing muscle soreness R
Enhances Physical Performance: Some studies suggest L-Carnitine may benefit VO2max, muscle contraction, and glycogen sparing. R
Improves Insulin Sensitivity: L-Carnitine has been shown in several human studies to improve insulin resistance, a key hallmark of diabetes. It works by stimulating whole-body glucose uptake and oxidation. The mechanism appears to involve activation of the PDH complex, shifting cellular metabolism towards glucose utilization, and reducing lactate production due to improved pyruvate oxidation. R R
Safety: The major problem with oral L-Carnitine supplements is that its metabolism in the gut leads to the formation of TMAO, a cardiotoxic & carcinogenic compound. More on this in the explanation below. R
L-Carnitine may also negatively affect the thyroid at high dosages. Avoid supplementing if you have weak thyroid function. R
My Dosage: I typically inject 500mg every other day of Injectable L-Carnitine as part of my pre-workout stack. Works best when taken with carbohydrates. I have friends who use up to 900mg EOD.
Where to Buy: You can get Injectable L-Carnitine from Amino Asylum or with a prescription from Defy Medical.
Why I’m Injecting It
Oral L-Carnitine L-Tartrate definitely works as well. The main problem with oral L-Carnitine is the formation of TMAO in your gut. It’s carcinogenic & cardiotoxic, but this can largely be mitigated with a garlic extract supplement. Note this extract must be rich in allicin, which is the key bioactive we need to counteract the TMAO. AlliMax is a great product you can use. The typical dose is about 5g (and at this dose, I would definitely add a garlic supplement). R
Side note, but Alpha GPC also generates TMAO, which is why I recommend other choline sources like Citicoline for daily use and reserve Alpha GPC for intense mental activity. R
To a lesser extent, Acetyl-L-Carnitine (aka ALCAR) is also efficacious, but is better suited for cognitive purposes due to its “acetyl” group, which allows it to freely cross the blood-brain barrier and get to work on the brain. It’s likely safer than L-Tartrate as it’s used in lower doses (500-2000mg) and probably doesn’t generate as much TMAO, if any. There aren’t any studies that prove that ALCAR increases TMAO and interestingly enough, there are studies showing ALCAR is actually cardioprotective rather than cardiotoxic. R R
2) Tirzepatide
Tirzepatide is the most effective appetite suppressant I’ve ever used.
In fact, it’s so powerful I could only take it for a few weeks before I had to stop; I was losing too much weight at the lowest effective dose (2.5mg/week).
Ozempic on Steroids
For those who don’t know, Tirzepatide is essentially an updated & more effective version of Ozempic (known generically as semaglutide).
Like Ozempic, it activates GLP-1 signaling obliterates your appetite.
But Tirzepatide actually goes one step further and activates GIP signaling as well. In fact, it’s actually more of a GIP receptor agonist than a GLP-1 receptor agonist.
For this reason, it’s actually much more effective than Ozempic inducing weight loss.
All in all, Tirzepatide:
Reduces blood sugar levels
Increases insulin release
Increases insulin sensitivity
Stimulates thermogenesis R
Reduces appetite
Slows gastric motility
Safety: You should consult an intelligent physician prior to using Tirzepatide (or any other drug, supplement, peptide, etc). Tirzepatide is currently approved by the FDA for the treatment of Type 2 Diabetes (as “Mounjaro”) & Obesity (as “Zepbound”). Side effects of Tirzepatide are generally mild at lower doses, such as nausea, constipation, diarrhea, and feeling full quickly, are similar to other GLP-1 receptor agonists and are considered manageable. Tirzepatide and similar drugs have also been linked to more severe side effects. I’d like to address 2 in particular below (note this does not include all potential severe side effects). These are not necessarily common, but possible.
Thyroid Cancer
In rats and mice, tirzepatide led to thyroid C-cell tumors and this risk likely applies to humans as well. The mechanism is thought to be related to the drug's action on GLP-1 receptors, which could stimulate thyroid C-cell proliferation and potentially lead to cancer.
Long-Lasting/Permanent Gastric Paralysis
Tirzepatide affects gastric motility; this is part of how it works to decrease appetite and make you feel “fuller”. This may exacerbate symptoms in individuals with pre-existing gastroparesis, which is sometimes an existing complication in diabetes/obesity. However, this slowing of gastric emptying is also a mechanism by which tirzepatide reduces postprandial glucose levels. There have been a few case reports of long-lasting gastric paralysis in those who take Tirzepatide, even after stopping the drug.
My Dosing: 2.5mg once per week. At this dose I noticed substantial appetite suppression without any significant nausea or GI symptoms. Clinical trials have used up to 15mg, but I personally would not recommend using more than 5mg once per week. Always consult a certified medical professional prior to using a potent tool like Tirzepatide.
Where to Buy: You can get N-Acetyl Tirzepatide Amidate from Limitless Life Nootropics for research purposes only (15% off with “nootbro”). You will likely need to sign up for a VIP account in order to buy it, which you can do here in order to purchase Tirzepatide and other newer peptides. N-Acetyl Tirzepatide Amidate is slightly more potent & longer-lasting than standard tirzepatide due to the acetyl & amidate group added to the peptide’s structure. This specific analogue has not been tested in human trials, but works as intended based on my experience employing it.
Side Note on Retatrutide: A new GLP-1 triple agonist has been becoming more popular over the past 6 months. I personally haven’t felt the need to use it as Tirzepatide works perfectly for me. It’s called Retatrutide and it stimulates not only GLP-1 & GIP receptors, but also Glucagon receptors. According to the initial research I’ve seen, Retatrutide is even more powerful at inducing weight loss than Tirzepatide. I don’t think this is necessary for most people though.
3) 5-Amino 1MQ
5-Amino 1MQ is the most exciting compound on this list.
It has the exceptional ability to powerfully burn fat & build muscle at the same time.
Let’s start with its effect on fat loss.
5-Amino 1MQ burns fat by upregulating cellular metabolism. It does this via inhibiting the NNMT enzyme, which is linked to both Type 2 Diabetes & Obesity.
Put simply, this enzyme slows down fat burning.
Inhibiting the NNMT enzyme:
Increases metabolic rate, glucose utilization/clearance, & insulin sensitivity via upregulating GLUT4 expression
Increases the longevity compound NAD+, which plays a role in cellular metabolism, muscle function, longevity, mitochondrial energy, & brain energy homeostasis
Increases SAMe, an important bioprotective & longevity compound
I’ve definitely noticed improved body composition over the course of 3 week cycles with 5A1MQ. I also felt noticeably more energized, probably due to the increase in NAD+ & SAMe.
Inhibiting NNMT expression is also anabolic.
Research & patents suggest that lowering NNMT expression can:
Promote the regeneration of aged muscle cells R
Repair muscle damage R
Improve neuromuscular function R
Enhance muscle fiber contraction potential R
Safety: 5-Amino 1MQ is an experimental small molecule drug (note: it’s not a peptide) with no formal human safety data. With that said, anecdotal experience reports & animal studies suggest it’s relatively safe. I personally didn’t notice any side effects from my 3 week cycles with 150mg of 5A1MQ.
My Dosage: I jumped the gun and ran 150mg per day (in the morning with breakfast) for 3 weeks. Due to its experimental nature, I wouldn’t recommend using it longer than this. It will also become less effective the longer you take it. I’d recommend taking it for 3 weeks at a time and taking 4-8 weeks off between cycles.
