Butea Superba

Nobody knows about Butea Superba. Yet.

It’s a traditional Thai herb that has been used for centuries to supercharge male vitality.

Modern human research on Butea Superba is limited. But the few trials & case studies that exist show Butea Superba is nothing to ignore.

One case study showed that Butea Superba supercharged one man’s DHT levels to supraphysiological levels (about 1.5x more than the top of the reference range) (R)

It’s very cheap and definitely worth a shot. I’ve used Swanson in the past which works.

2. Tribulus Terrestris

Tribulus is one of my all-time favorite herbs.

I ignored it for so long as many think it’s just another scam. This is because it’s marketed as a testosterone booster but does not necessarily boost testosterone levels.

But what it does do is supercharge DHT levels.

One human study showed that Men who took 250mg Tribulus Terrestris (TT) daily had a significant decrease in the percentage of body fat and an increase in lean mass as well as an increase in DHT levels (R).

Tribulus (112mg protodioscin) daily (R increased DHT by 27% and lowered prolactin by almost 60%.

I am currently using 1000mg Tribulus (20% Protodioscin) from Nootropics Depot as I’m writing this post.

This means I’m getting 200mg from Tribulus per day, which should be enough to boost DHT 20-40%+ based on the human trials results with 112.5mg protodioscin (boosted DHT by an average of 27%.

3. Boron

Boron is androgenic AF.

It’s a trace mineral that increases free testosterone and free DHT. This is because it’s a potent inhibitor of sex hormone binding globulin (SHBG).

Here’s the part 99% of gurus miss: SHBG binds to DHT with about 5x the affinity of testosterone.

This means that lowering SHBG with boron is going to have a MUCH more potent effect on free DHT than on free testosterone. And the studies show it already has a potent effect on free testosterone.

I use 6mg of elemental boron from Nootropice Depot’s Boron Glycinate Nootropics Depot

Pro Tip: Tongkat Ali obliterates SHBG similar to boron. One human study showed that it reduced SHBG by 66%, ultimately leading to a 73% increase in free testosterone (R). Free DHT wasn’t measured, but I’d predict it increased significantly both due to the increase in testosterone as well as reduction in SHBG.

Compounds in this post may be available for purchase for non-human research purposes only. We do not condone the use of research chemicals for unapproved purposes.

1. L-Carnitine

L-carnitine is just an amino acid, but it’s the FURTHEST thing from boring.

First, it turns fat into fuel.

More specifically, it works by transporting long-chain fatty acids into the mitochondria to be burned for energy by a process called β-oxidation. R

And because of this, it’s been proven in human studies to enhance weight loss, even when taken orally. R R

But L-Carnitine also has potent androgenic activity.

Specifically, it’s been shown to upregulate androgen receptors.

A rat study showed that L-carnitine-L-tartrate (LCLT) increased androgen receptors by 15% in 3 weeks.2

A human study actually showed that L-Carnitine can restore sexual function and is even more potent than testosterone at enhancing erections. L-Carnitine does this WITHOUT significantly altering any sex hormone levels.3

My Dosage: I typically inject 500mg every other day of Injectable L-Carnitine as part of my pre-workout stack. Works best when taken with carbohydrates. I have friends who use up to 900mg EOD. You can also use 1-5g of L-Carnitine Tartrate per day. You’ll want to read my Fat Loss substack in order to learn how to mitigate the cardiotoxicity associated with oral L-Carnitine.

Where to Buy: You can get Injectable L-Carnitine from Amino Asylum or with a prescription from Defy Medical. Otherwise, I’d just get L-Carnitine Tartrate off of Amazon.

2. Tribulus

Tribulus is my favorite androgenic herb. A human study4 showed that it can:

Increase DHT by 27%

Increase DHEA by 183%

Decrease prolactin by 56%

There are virtually no other supplements that can do all of the above to the same extent. But its benefits don’t stop there. A rat study showed it can upregulate androgen receptors by 58%.5

The additional neural androgen receptors combined with higher levels of DHT is likely why Tribulus is the most potent androgenic herb for confidence, aggression, and motivation.

Tribulus likely increases androgen receptors by increasing DHT. When DHT (and testosterone) activate androgen receptors, the downstream result is actually more synthesis of androgen receptors. This is in contrast to higher levels of dopamine which will downregulate dopamine receptors.

Higher levels of testosterone + DHT + DHEA → More Activation of Androgen Receptors → More Downstream Transcription → More Synthesis of Androgen Receptors

Important note: the human study used a product containing 112.5mg of protodioscin. This is the secret sauce in Tribulus that has been shown to upregulate the 5AR enzyme, and thus DHT production. This means that 1 capsule of Nootropics Depot’s Tribulus product will have a significant effect on DHT.

You can get their Tribulus here (10% off with NOOTBRO10). I recommend taking 1-2 capsules daily with 250mg of TUDCA to protect the liver.

It’s just a boring amino acid right?

Wrong.

It’s quickly becoming one of my favorite nutrients of all-time the more I use it. This became even more apparent when I switched over from L-Carnitine L-Tartrate & ALCAR to Injectable L-Carnitine. More on why I did that later.

The #1 reason why I use it?

It turns fat into fuel.

More specifically, it works by transporting long-chain fatty acids into the mitochondria to be burned for energy by a process called β-oxidation. R

And because of this, it’s been proven in human studies to enhance weight loss, even when taken orally. R R

L-Carnitine’s fat-burning effects are even more prominent when used in conjunction with medium to low exertion cardio through its lipid oxidisation and glycogen sparing abilities R

Additional Relevant L-Carnitine Benefits

Androgenic Activity: L-Carnitine intake has also been shown to upregulate androgen receptors, which basically makes your testosterone work harder for you. All of the androgenic benefits you normally get from testosterone will be enhanced while using taking L-carnitine, including a massive benefit sexual performance. It’s so powerful that it has been called a natural alternative to TRT. And in a human trial, L-Carnitine actually outperformed TRT in some markers of sexual function without significantly altering any sex hormone levels. When I started taking higher doses of L-Carnitine, I definitely noticed improved sexual health as well (stronger erections & sex drive mainly). Improved androgen signaling will also favorably benefit body composition. R R

Increases IGF-1 levels: IGF-1 is the anabolic hormone responsible for many of the hypertrophic benefits of growth hormone. R

Increases ATP production: By turning fat into fuel, L-Carnitine also increases mitochondrial ATP energy output, resulting in a noticeable energy boost which is especially useful for workouts. R

Enhances Muscle Recovery: L-Carnitine has been shown to improve blood flow, oxygen, & glycogen levels in muscles. This is in part why it expedites muscle recovery and reducing muscle soreness R

Enhances Physical Performance: Some studies suggest L-Carnitine may benefit VO2max, muscle contraction, and glycogen sparing. R

Improves Insulin Sensitivity: L-Carnitine has been shown in several human studies to improve insulin resistance, a key hallmark of diabetes. It works by stimulating whole-body glucose uptake and oxidation. The mechanism appears to involve activation of the PDH complex, shifting cellular metabolism towards glucose utilization, and reducing lactate production due to improved pyruvate oxidation. R R

Safety: The major problem with oral L-Carnitine supplements is that its metabolism in the gut leads to the formation of TMAO, a cardiotoxic & carcinogenic compound. More on this in the explanation below. R

L-Carnitine may also negatively affect the thyroid at high dosages. Avoid supplementing if you have weak thyroid function. R

My Dosage: I typically inject 500mg every other day of Injectable L-Carnitine as part of my pre-workout stack. The main problem with oral L-Carnitine is the formation of TMAO in your gut. It’s carcinogenic & cardiotoxic, but this can largely be mitigated with a garlic extract supplement. Note this extract must be rich in allicin, which is the key bioactive we need to counteract the TMAO. AlliMax is a great product you can use. The typical dose is about 5g (and at this dose, I would definitely add a garlic supplement). R

Side note, but Alpha GPC also generates TMAO, which is why I recommend other choline sources like Citicoline for daily use and reserve Alpha GPC for intense mental activity. R

To a lesser extent, Acetyl-L-Carnitine (aka ALCAR) is also efficacious, but is better suited for cognitive purposes due to its “acetyl” group, which allows it to freely cross the blood-brain barrier and get to work on the brain. It’s likely safer than L-Tartrate as it’s used in lower doses (500-2000mg) and probably doesn’t generate as much TMAO, if any. There aren’t any studies that prove that ALCAR increases TMAO and interestingly enough, there are studies showing ALCAR is actually cardioprotective rather than cardiotoxic. <a release

Increases insulin sensitivity

Stimulates thermogenesis R

Reduces appetite

Slows gastric motility

Safety: You should consult an intelligent physician prior to using Tirzepatide (or any other drug, supplement, peptide, etc). Tirzepatide is currently approved by the FDA for the treatment of Type 2 Diabetes (as “Mounjaro”) & Obesity (as “Zepbound”). Side effects of Tirzepatide are generally mild at lower doses, such as nausea, constipation, diarrhea, and feeling full quickly, are similar to other GLP-1 receptor agonists and are considered manageable. Tirzepatide and similar drugs have also been linked to more severe side effects. I’d like to address 2 in particular below (note this does not include all potential severe side effects). These are not necessarily common, but possible.

Thyroid Cancer

In rats and mice, tirzepatide led to thyroid C-cell tumors and this risk likely applies to humans as well. The mechanism is thought to be related to the drug's action on GLP-1 receptors, which could stimulate thyroid C-cell proliferation and potentially lead to cancer.

Long-Lasting/Permanent Gastric Paralysis

Tirzepatide affects gastric motility; this is part of how it works to decrease appetite and make you feel “fuller”. This may exacerbate symptoms in individuals with pre-existing gastroparesis, which is sometimes an existing complication in diabetes/obesity. However, this slowing of gastric emptying is also a mechanism by which tirzepatide reduces postprandial glucose levels. There have been a few case reports of long-lasting gastric paralysis in those who take Tirzepatide, even after stopping the drug.

My Dosing: 2.5mg once per week. At this dose I noticed substantial appetite suppression without any significant nausea or GI symptoms. Clinical trials have used up to 15mg, but I personally would not recommend using more than 5mg once per week. Always consult a certified medical professional prior to using a potent tool like Tirzepatide.

Where to Buy: You can get N-Acetyl Tirzepatide Amidate from Limitless Life Nootropics for research purposes only (15% off with “nootbro”). You will likely need to sign up for a VIP account in order to buy it, which you can do here in order to purchase Tirzepatide and other newer peptides. N-Acetyl Tirzepatide Amidate is slightly more potent & longer-lasting than standard tirzepatide due to the acetyl & amidate group added to the peptide’s structure. This specific analogue has not been tested in human trials, but works as intended based on my experience employing it.

Side Note on Retatrutide: A new GLP-1 triple agonist has been becoming more popular over the past 6 months. I personally haven’t felt the need to use it as Tirzepatide works perfectly for me. It’s called Retatrutide and it stimulates not only GLP-1 & GIP receptors, but also Glucagon receptors. According to the initial research I’ve seen, Retatrutide is even more powerful at inducing weight loss than Tirzepatide. I don’t think this is necessary for most people though.

3) 5-Amino 1MQ

<picture></picture>

<g>

</g><polyline points="15 3 21 3 21 9"></polyline><polyline points="9 21 3 21 3 15"></polyline><line x1="21" x2="14" y1="3" y2="10"></line><line x1="3" x2="10" y1="21" y2="14"></line>

5-Amino 1MQ is the most exciting compound on this list.

It has the exceptional ability to powerfully burn fat & build muscle at the same time.

Let’s start with its effect on fat loss.

5-Amino 1MQ burns fat by upregulating cellular metabolism. It does this via inhibiting the NNMT enzyme, which is linked to both Type 2 Diabetes & Obesity.

Put simply, this enzyme slows down fat burning.

Inhibiting the NNMT enzyme:

Increases metabolic rate, glucose utilization/clearance, & insulin sensitivity via upregulating GLUT4 expression

Increases the longevity compound NAD+, which plays a role in cellular metabolism, muscle function, longevity, mitochondrial energy, & brain energy homeostasis

Increases SAMe, an important bioprotective & longevity compound

I’ve definitely noticed improved body composition over the course of 3 week cycles with 5A1MQ. I also felt noticeably more energized, probably due to the increase in NAD+ & SAMe.

Inhibiting NNMT expression is also anabolic.

Research & patents suggest that lowering NNMT expression can:

Promote the regeneration of aged muscle cells R

Repair muscle damage R

Improve neuromuscular function R

Enhance muscle fiber contraction potential R

Safety: 5-Amino 1MQ is an experimental small molecule drug (note: it’s not a peptide) with no formal human safety data. With that said, anecdotal experience reports & animal studies suggest it’s relatively safe. I personally didn’t notice any side effects from my 3 week cycles with 150mg of 5A1MQ.

My Dosage: I jumped the gun and ran 150mg per day (in the morning with breakfast) for 3 weeks. Due to its experimental nature, I wouldn’t recommend using it longer than this. It will also become less effective the longer you take it. I’d recommend taking it for 3 weeks at a time and taking 4-8 weeks off between cycles.

Where to Buy: You can get 5-Amino 1MQ for research purposes from Limitless Life Nootropics. This stack is designed for performance & longevity to keep me at the top of my game both today & in 30 years.

This protocol will be split into 4 parts: general health supplements, testosterone boosters & aphrodisiacs, nootropics, & drugs. This reflects what I consistently take daily. This does not include as-needed agents or other compounds I’m experimenting with.

Let’s get started…

MEDICAL DISCLAIMER

The NootBro newsletter does not contain medical advice. Although I make efforts to keep the medical information in this newsletter current, I cannot guarantee that the information in this newsletter or on my social media channels reflects the most up-to-date research.

Consult your physician for medical advice. Always seek the advice of a physician or other qualified healthcare provider with any questions regarding a medical condition. Never disregard or delay seeking professional medical advice or treatment because of something you have heard on the Internet. Before taking any medications, over-the-counter drugs, supplements, or herbs, consult a physician for a thorough evaluation. This channel does not endorse any medications, vitamins, or herbs nor do we condone the use of illegal drugs or using drugs for an unintended purpose. A qualified physician should make a decision based on each person's medical history and current prescriptions.

General Health Supplements

#1 - N-Acetyl Cysteine (NAC)

N-Acetyl Cysteine, or NAC, is a potent antioxidant & glutathione precursor that helps to increase the levels of glutathione in the body. I take NAC for the antioxidant benefits, liver protection, & brain benefits. NAC is also especially helpful for psychiatric disorders such as OCD (Source), which I used to struggle with. I typically take 1200mg per day of Nootropics Depot’s NAC.

#2 - TUDCA

TUDCA, or Tauroursodeoxycholic acid, is a bile acid that can be naturally found in the body. I take TUDCA to protect & enhance my liver function (Source). I do this out of an abundance of caution, as I consume many herbal supplements that may have some degree of hepatotoxicity. I typically take 500mg per day of Nutracost’s TUDCA.

#3 - Astragalus

Astragalus is a traditional Chinese herb that has been shown to have potent anti-aging properties. I take Astragalus to support my kidney health and protect them from damage caused by oxidative stress (Source). Studies have also shown that Astragalus can promote telomere elongation, which can improve cellular health and slow down the aging process. I take 5g of Astragalus Extract from NOW per day.

#4 - Citrus bergamot

Citrus bergamot is one of the best, if not the best, supplements to lower cholesterol (Source). Citrus Bergamot is also a strong antioxidant that can help to reduce oxidative stress in the body. It has also been shown to inhibit PCSK9 expression, a protein that can interfere with the liver's ability to remove LDL cholesterol from the bloodstream. By inhibiting PCSK9, Citrus Bergamot can help to further lower LDL cholesterol levels and reduce the risk of heart disease. I use one serving of U.S. Doctors’ Clinical CitriCholess® as it is the form that has been used in clinical trials.

#5 - Nigella sativa

Nigella Sativa, also known as Black Seed or Black Seed Oil, is a widely used medicinal plant with a long history of use in traditional medicine systems. According to Prophet Muhammed, it’s “the cure for everything except death.” While this is a bit of a stretch, the science seems to agree with him:

A review on therapeutic potential of Nigella sativa: A miracle herb

Revisiting pharmacological potentials of Nigella sativa seed: A promising option for COVID-19 prevention and cure

An updated literature-based review: phytochemistry, pharmacology and therapeutic promises of Nigella sativa

I take nigella sativa primarily to enhance immune function & reduce systemic inflammation, but it also has a wide range of other therapeutic properties, including antidiabetic, anticancer, analgesic, antimicrobial, anti-inflammatory, spasmolytic, bronchodilator, hepato-protective, renal protective, gastro-protective, anti-depressant, anxiolytic, and antioxidant properties. The primary bioactive component of Nigella Sativa is thymoquinone, which is found in the essential oil of the seeds. If you ever start taking nigella sativa or black seed oil, I recommend finding an extract/oil with standardized to at least 3-5% of thymoquinone content. I take 400mg of nigella sativa extract (5% thymoquinone) from Nootropics Depot daily.

#6 - Sodium Butyrate

Butyrate is a short-chain fatty acid that is produced by the gut microbiota and is also found in dietary sources like butter and cheese. I take Sodium Butyrate (as this is the supplemental form used in many studies) primarily for its potent gut-healing properties. It activates the GPR109A receptor which influences intestinal permeability & gastrointestinal inflammation. It also has anti-cancer properties via HDAC inhibition & neuroprotective properties via inducing BDNF & GDNF. I take 2 capsules (equivalent to 1.2g butyric acid) per day from BodyBio.

#7 - Goat Colostrum

Goat colostrum is a superfood that contains a rich array of active immune systems compounds, such as peptides, immunoglobulins (IgG, IgA, and IgM), lactoferrin, cytokines, and more than 20 different antibodies for immune support. Lactoferrin is particularly important when trying to control unbound iron, which is highly inflammatory and contributes to all sorts of issues. That said, I take goat colostrum primarily to bulletproof my digestive system & reduce intestinal permeability (otherwise known as “leaky gut”). I take 1 serving of Mt. Capra’s goat colostrum daily.

By the way, goat colostrum is thought to be more easily digestible than bovine colostrum and absorbed by the body due to its smaller fat globules and looser protein curd structure. This may make it a more suitable option for individuals with lactose intolerance, cow's milk allergies, or sensitive digestive systems.

#8 - Liposomal Glutathione

Liposomal Glutathione is a highly bioavailable form of the "master antioxidant" with approximately 10x the absorption of traditional glutathione supplements. Its potency lies in its ability to combat oxidative stress, neutralize harmful free radicals, & reduce inflammation. In turn, this protects your body & allows your cells to function optimally. The corresponding benefits are almost countless:

enhance detoxification

improve skin aesthetics

protect brain

reduce mental illness

enhance immunity & fight infections

improve cardiovascular health

improve gut health

In other words, it does just about everything. Rho Nutrition has very high-quality liposomal glutathione that I use 1-2 servings of daily. You can use the code ‘NOOTBRO10” to get 10% off.

Testosterone & Aphrodisiac Supplements

#9 - Shilajit

Shilajit is a naturally occurring resin that has been used for thousands of years in indigenous medicine systems for its anti-inflammatory, antioxidant, aphrodisiac, anti-pain, mitochondrial enhancing, and nootropic activities. Its benefits are largely attributed to the high concentration of fulvic & humic acids and dibenzo-α-pyrones, as well as its mineral makeup. I specifically use Shilajit to boost testosterone, libido, & energy. It’s one of the few herbal supplements that you can really “feel” despite it not having a direct action on dopamine or norepinephrine. I take 8 tablets of Based Shilajit dissolved in spring water daily. I don’t recommend taking 8 though; 2-4 does the trick and is less likely to cause any side effects. I mix mine in 1L of spring water bottled. in glass.

Shilajit is a supplement that often is contaminated with heavy metals & mycotoxins, so you have to be careful who you buy from. Based does extensive third-party lab testing to confirm purity, potency, & lack of contaminants for their products. You can use ‘NOOTBRO’ for 10% off.

#10 - Cistanche tubulosa

Cistanche is a herb commonly used in traditional Chinese medicine for its potential health benefits, including positively modulating the gut-brain axis, increasing expression of GDNF for dopaminergic neuronal repair, and improving testosterone levels and fertility by enhancing steroidogenic enzyme expression. I take 200mg of Cistanche tubulosa extract from Nootropics Depot daily.

#11 - Tribulus

Tribulus Terrestris is a herb commonly used for its potential benefits in increasing testosterone levels and enhancing athletic performance. It contains steroidal saponins called protodioscin and dioscin, which have been shown to increase DHT production by increasing the 5-alpha-reductase enzyme. Additionally, it contains beta-carbolines that inhibit MAO enzymes, resulting in greater levels of serotonin, dopamine, and norepinephrine. I take 500mg of Tribulus from Nootropics Depot daily.

Note that Tribulus may raise liver enzymes, but taking TUDCA & NAC will help mitigate this damage. High doses of astragalus extract will help counteract any potential kidney damage from Tribulus or other herbal supplements.

Nootropics

#12 - Notable Focus

Notable Focus is a formulation designed to enhance cognition quickly using underrated herbal ingredients. I won’t be revealing the entire formula here, but it contains two heavy hitters: Sabroxy & Polygala tenuifolia. Both of these have been shown to boost dopamine (& other neuromodulators) significantly in a way that vastly outperforms almost all other supplements, nootropics, & herbs. Both inhibit monoamine reuptake, and Poylgala also inhibits monoamine oxidase, resulting in significantly higher levels of dopamine. I take 1-2 servings per day.

Sabroxy has been shown to significantly enhance motivation & focus in human studies. This is due to Oroxylin A’s ability to inhibit dopamine reuptake, leading to greater concentrations of dopamine in the brain (Source). Oroxylin A also increases brain-derived neurotrophic factor (BDNF) in the brain, which increases neurogenesis, protects against neurotoxicity, & enhances memory (Source). In addition to its powerful cognitive-enhancing benefits, Sabroxy can also help reduce inflammation, improve skin health, and strengthen the immune system by inhibiting the COX-2 gene (Source). I recommend taking 300 to 500mg as needed with days off to prevent tolerance buildup.

Recommended Product: Sabroxy

Polygala Tenuifolia

Polygala Tenuifolia, also known as Yuan zhi, is an herb from traditional Chinese practices. Recently, Polygala Tenuifolia has become the subject of academic & scientific research due to its cognitive-enhancing capabilities. Polygala Tenufolia supports motivation & focus by inhibiting monoamine reuptake (Source) & inhibiting monoamine oxidase (Source). This increases concentrations of dopamine, norepinephrine, & serotonin in the brain. Polygala also enhances memory by inhibiting acetylcholinesterase, the enzyme that breaks down acetylcholine, a neurotransmitter associated with learning & memory (Source). Polygala also increases BDNF & nerve growth factor (NGF) expression which further benefits memory and contributes to Polygala’s neurogenic & neuroprotective effects. (Source) I recommend taking 100mg of Polygala 20:1 up to 3x daily.

Recommended Product: Polygala 20:1
